Login failed for user '<username>'. Reason: Failed to open the explicitly specified database 'BarTender'. [CLIENT: <local machine>]
BarTender 2016 R8. Have not setup a MSSQL$BARTENDER database but BarTender is trying to login into this SQL Express database which was never created or configured in the Administration Console. Is there a registry setting which needs to be updated or deleted in order to stop these information logs being created in the Application Log of Event Viewer

I believe this error you are experiencing is related to the connection to the database inside the label, rather than a connection to the SQL Express Database (which as you said, wasn't even created).
I would suggest opening your working labels with BarTender Designer and checking whether they can successfully connect to the databases they are connected with. If not, please reconnect them to their databases accessing the Database Setup menu on them and selecting "Configure Connection" on their properties connection screen:

We are getting a the same issue at one of our customers sites. They are using version 2021 R8. So the messages say 'Datastore' instead of 'Bartender'
The label formats we have aren't connected to a database. We populate the data on the label directly using named datasources.
The customer is complaining about these messages going in to the windows event viewer.
I've turned off all the logging I can see in the Administrator Console.
Is there a way to stop these messages from being logged?
