I take you are using either the Automation or Enterprise edition and have checked that the Integration Service is running.

Have you tested the Integration file you created and Integration Builder and did not receive any errors when you tested it?

If so, then click on the Deployment tab and use the New Deployment option to manually configure and start the integration and see if that works, rather than deploying the integration from Integration Builder.

I have the same issue and get this message, how to fix this?

ERROR|Loading deployment file failed. Details: An error occurred accessing the database. This usually means that the connection to the database failed. Check that the connection string is correct and that the appropriate DbContext constructor is being used to specify it or find it in the application's config file. See http://go.microsoft.com/fwlink/?LinkId=386386 for information on DbContext and connections. See the inner exception for details of the failure. The provider did not return a ProviderManifestToken string. A network-related or instance-specific error occurred while establishing a connection to SQL Server. The server was not found or was not accessible. Verify that the instance name is correct and that SQL Server is configured to allow remote connections. (provider: SQL Network Interfaces, error: 50 - Local Database Runtime error occurred. Cannot create an automatic instance. See the Windows Application event log for error details.«CR»«LF»)

It sounds look you are having a problem connecting to the system database. Are all the services running? Did you use the standard Database config or a custom on?
